Role of Synovial Exosomes in Osteoclast Differentiation in Inflammatory Arthritis

چکیده
This study aimed to investigate the characteristics of exosomes isolated from synovial fluid and their role in osteoclast differentiation different types inflammatory arthritis. Exosomes rheumatoid arthritis (RA), ankylosing spondylitis (AS), gout, osteoarthritis (OA) patients were co-incubated with CD14+ mononuclear cells healthy donors without macrophage colony-stimulating factor (M-CSF) receptor activator nuclear kappa-B ligand (RANKL). Osteoclast was evaluated via tartrate-resistant acid phosphatase (TRAP) staining activity F-actin ring formation. RANKL expression on assessed using flow cytometry an enzyme-linked immunosorbent assay (ELISA). Synovial lowest OA patients; these induced osteoclastogenesis absence M-CSF RANKL. Osteoclastogenesis significantly higher more RA (p = 0.030) than patients, but not AS or gout patients. On treating macrophages a specified number RA/AS greater exosomal levels 0.035) exosome numbers vary type may bear disease-specific “synovial signature osteoclastogenesis.”

BACKGROUND Pathological bone resorption (marginal erosions and juxta-articular osteoporosis) by osteoclasts commonly occurs in rheumatoid arthritis (RA). OBJECTIVES To define the nature of the mononuclear precursor cells from which osteoclasts are formed in inflamed synovial tissues and to determine the cellular and humoral factors which influence osteoclast differentiation. METHOD Macropha...
متن کاملrole of cultural iran in promotion of multilaieralism in central asia and caucasus
cultural iran is a scope that is more extended than the political territories of iran as a political unit. this concept means that cultural geography(mehdi moghanlo-1383-1) of iran is greater than its political geography which, according to history, has a long history extending west-east from kandahar to the euphrates and north-south from the persian gulf to the caucasus including transoxiana a...

Bone disease is the most frequent complication in multiple myeloma (MM) resulting in osteolytic lesions, bone pain, hypercalcemia and renal failure. In MM bone disease the perfect balance between bone-resorbing osteoclasts (OCs) and bone-forming osteoblasts (OBs) activity is lost in favour of OCs, thus resulting in skeletal disorders. Since exosomes have been described for their functional role...
متن کاملSynovial tissue in rheumatoid arthritis is a source of osteoclast differentiation factor.
OBJECTIVE Osteoclast differentiation factor (ODF; also known as osteoprotegerin ligand, receptor activator of nuclear factor kappaB ligand, and tumor necrosis factor-related activation-induced cytokine) is a recently described cytokine known to be critical in inducing the differentiation of cells of the monocyte/macrophage lineage into osteoclasts. The role of osteoclasts in bone erosion in rhe...
